Carrot cells (Daucus carota L.) cultured in an IAA and m-inositol containing medium were able to produce adventitious roots and somatic embryos. These differentiational processes could be suppressed by a kinetin supplement, which also increases cell proliferation. In synchronized carrot cell suspensions (using a Fluordesoxyuridin/Thymidine system) in the plus kinetin treatment soon after thymidine application the cells passed into the S-phase whereas in the kinetin free nutrient medium the entrance into the S-phase was delayed for about 1 hour during which the cells remained in the G1-phase. During this lag period some 140 proteins are synthesized in a hierachical sequence (C-14-leucine as a tracer), which were missing in the plus kinetin cultures. It remains to be seen to which extent these additional proteins synthesized during the prolonged G1-phase contribute to the ability of the cells of this treatment to enter the S-phase later and/or the ability of these cultures to differentiate adventitious roots or somatic embryos.
