Isolation of intact storage tissue amyloplasts from suspension-cultured potato cells (Solanum tuberosum) and determination of their intermembrane and stroma volumes

Abstract
Storage tissue amyloplasts were isolated from protoplasts of a heterotrophic cell suspension culture of potato (Solanum tuberosum L., dihaploid stock, HH258) in the physiological phase of the beginning of starch synthesis. Protoplasts were obtained after a short (90 min) enzyme (Cellulase Onozuka RS and Pectolyase Y-23) incubation of the potato cells. A homogenous fraction of amyloplasts was prepared from ruptured protoplasts by centrifugation (100 g for 15 min) through a discontinuous Ficoll density gradient. Amyloplasts having an uniform diameter of 5 mu m were obtained at the interphase between 10 % and 25 % -Ficoll. About half of these isolated amyloplasts were intact and there was only a small degree (0 - 1.5 %) of contamination by other cellular compartments (KOSEGARTEN and MENGEL, 1994). The extensions of the free diffusible water space and of the space accessible to sucrose were estimated by means of H-3 labelled water and C-14 labelled sucrose. The free diffusible space of the amyloplasts comprised a volume of about 10 % of the entire amyloplast which means that about 90 % of the amyloplast volume was filled with starch. The proportion of the stroma space of these storage tissue amyloplasts was about 28 % of the free diffusible space and is thus smaller than in chloroplasts, etioplasts and root amyloplasts.
