Abstract:
The cellular targets of adriamycin (ADR) action were described in human mesangial cell (HMC) cultures and Tetrahymena thermophila by a comparative study of growth and biomass production in the presence of the drug (0.5 to 20 mug/ml). In both cell lines cell division was arrested dose-dependently, accompanied by cell volume increase up to a factor of 2. Inhibition of culture growth was correlated with an inhibition of RNA synthesis as revealed by precursor incorporation studies. RNA quality was, however, unaffected as indicated by the sedimentation profiles of total RNA. The inhibitory effect on protein synthesis in HMC and Tetrahymena was weak and delayed and therefore regarded as secondary. DNA synthesis was decreased dose-dependently in HMC cultures. It was unaffected in Tetrahymena by high ADR concentrations while it was stimulated by low doses. The rates of biomass accumulation in Tetrahymena substantiated the precursor incorporation studies and further confirmed a primary inhibitory effect of ADR on RNA production. Inhibition of growth and biomass production in Tetrahymena was completely reversible upon ADR removal. The data presented are discussed with special reference to Tetrahymena as a suited test organism.
