Abstract:
Extracts from ground leaves of Melia azedarach L. inhibit growth and disrupt metamorphosis of the fourth instar larvae of the Mexican bean beetle, Epilachna varivestis Muls. A biologically active principle was isolated by open column liquid chromatography (LC), preparative thin layer chromatography (TLC) and preparative high performance liquid chromatography (HPLC). Analytical TLC (chloroform - methanol 19:1) resulted in a single spot with a R(f) value of 0.35. Analytical HPLC on a RP-18 column (Lichrospher 5-mu-m, 250 x 4 mm) with acetonitrile and water (45 % : 55 %) as a mobile phase showed a retention time of the substrance of 5.0 min. UV-scanning revealed two maxima at 210 nm and 280 nm. A 100 % larval mortality was observed with the pure substance below 2 ppm in the bioassay.
