Abstract
Palytoxin caused within min a temperature-dependent K+ loss from human and rat erythrocytes which was followed within h by hemolysis. It decreased the osmotic resistance in a concentration-dependent manner, so that osmotic influences were negligible for K+ release but considerable in hemolysis. External K+ inhibited the Hb release and Rb+ inhibited the release of K+ and Hb. Ca2+ (> 20 .mu.M) and borate (> 5 .mu.M) enhanced the loss of K+ and Hb. With Ca2+ and borate present, the efficacy of palytoxin was raised .apprx. 10,000-fold. Under these conditions, .apprx. 15 palytoxin molecules/human cell triggered a 50% K+ loss over a wide range of cell concentrations. The palytoxin effect was reversible. After depletion from K+ by low concentrations of palytoxin, human cells could be refilled with K+ and resealed. The pores formed by palytoxin were small. They allowed the entrance of Na+ and choline; inositol was largely excluded and Ca2+, sucrose and inulin were excluded. Amphotericin B resembled palytoxin in its ability to cause a considerable prelytic K+ loss and to form small pores. It was .apprx. 1000-fold weaker than palytoxin, was not inhibited by K+ or Rb+, was not activated by Ca2+ or borate and had a negative temperature dependence. Palytoxin represented a novel type of cytolysin.
