Abstract
Application of electron diffraction to thin-sectioned intracellular crystalline inclusions using specimen tilting in the microscope is illustrated in monoclinic, orthorhombic and trigonal (rhombohedral) examples of protein crystals. Electron diffraction is more efficient than optical diffraction because of its applicability during microscopic study of the specimen and easy calibration. Interpretation of the diffraction patterns with respect to lattice symmetry and further analysis of the specimen [amphibian and teleost cyclostome yolk protein] is described.
