Abstract:
The energy profile of the proline biosynthetic sequence from Phaseolus vulgeris was calculated. This results in a .DELTA.G.degree.''[Gibbs free energy]-value of -55.4 kJ/mol for the overall reaction. The mass action ratios for the in vivo system must be determined. The equilibrium constant for the last step of the reaction sequence was evaluated experimentally. For the .DELTA.-pyrroline-5-carboxylate reductase reaction a value of 2.2 .times. 10-14 mol/l was found for the pH-independent constant. From the rate constants and the reversibility of this reaction it is assumed that the proline dehydrogenase in fact is a .DELTA.-pyrroline-4-carboylate reductase (E.C. 1.5.1.2.) Proline oxidation may proceed via a different reaction than the synthesis. Fundamental knowledge needed to understand the mechanism of proline accumulation under stress conditions is still missing.
