Abstract:
Palytoxin is a hemolysin. The erythrocytes from various species can be classified into a sensitive and a hardly sensitive group. The former contained K as its main inside cation and the erythrocytes were arranged according to their sensitivity as hog .gtoreq. rat, mouse > rabbit > guinea-pig > man. The latter, comprising those from sheep and cattle, had Na as its main inside cation. Chicken erythrocytes were relatively insensitive. Hemolysis of rat erythrocytes was preceded by a lag period of 1-2 h. With increasing temperature the hemolysis proceeded more quickly but reached the same final range between 25-42.degree. C. The pH optimum in Britton-Robinson buffer supplemented with saline was between 7-8. Washing off palytoxin during the prelytic period reduced the hemolytic power. The sensitivity of rat erythrocytes decreased with increased osmolarity between 235-415 mosM [milliosmolar]. Their osmotic resistance was lowered by palytoxin in a concentration-dependent manner. With rat and sheep erythrocytes, K loss preceded the hemolysis due to palytoxin. K loss was measurable already after 1 min and increased with time. After 2 h the quotient between the ED50 of hemolysis and that of K loss was .apprx. 200. Palytoxin was an unusually strong but slow hemolysin of the osmotic type. The extreme prelytic K loss and the correlation between susceptibility and K content of erythrocytes pointed towards the relevance of ionic fluxes.
