Abstract
A microfluorometric method of determining the relative DNA content in plant cells is described. Examples are given showing how ploidy levels in plants can be determined by this method. Standardization was achieved by using microspores in tetrads of the appropriate plant species. A comparison of n and 2n Datura innoxia plants showed that even in plants that were 30 wk old the majority of cells in the shoot axis of n plants are still haploid, whereas in 2n plants greater diversity of C-levels (up to 16 C) was found.
