CHROMATOGRAPHIC BEHAVIOR OF VIROIDS OF EXOCORTIS DISEASE OF CITRUS AND OF SPINDLE TUBER DISEASE OF POTATO

Abstract
The behavior of the viroid of the exocortis disease of citrus (ExCV) and that of the spindle tuber disease of potato (PSTV) was studied on different systems of column chromatography, including molecular sieving and adsorption chromatography. Total nucleic acid was extracted from viroid-infected plant tissue with the aid of phenol, purified partially, fractionated and analyzed by polyacrylamide gel electrophoresis. The viroid-containing nucleic acid preparations were analyzed by column chromatography and the presence of viroid-RNA in the different fractions was determined by bioassay. In molecular sieving on Sephadex G100 and Bio-Gel P200 viroid-RNA is eluted together with DNA and RNA of high molecular weight. On hydroxyapatite viroid-RNA elutes along with different cellular nucleic acids like a single-stranded molecule. Elution from CF 11-cellulose indicates the existence of a single-stranded and a double-stranded form of viroid-RNA. On DEAE-Sephadex viroid-RNA elutes with cellular 4S to 7S-RNA. Polysaccharides and polyphenols, which are always extracted together with plant nucleic acids can only be removed by molecular sieving. Final purification of viroid-RNA can be achieved by different combinations of columnchromatography followed by preparative electrophoresis in polyacrylamide gels.
