Abstract:
In a previous study, a high dosage of lifibrol (20 mg/kg) was shown to lower serum cholesterol in pigs. The present study was performed to investigate the effect of lower dosages of lifibrol (5 mg/kg and 10 mg/kg) on concentrations of serum lipids in the pig. 14 pigs were randomly divided into three groups that each received a diet without lifibrol, a diet with 5 mg/kg lifibrol or a diet with 10 mg/kg lifibrol. The diets were administered for 3 weeks. Blood samples were taken at the start of the experiment, during the experiment, and 4 days after the application of lifibrol had been stopped. At the end of the experiment, total serum cholesterol was 19% and 39% lower than at the beginning of the experiment in the groups treated with 5 mg/kg and 10 mg/kg lifibrol, respectively; non-esterified cholesterol was 33% and 49% lower, HDL cholesterol 37% and 35% lower. LDL cholesterol was markedly reduced by application of 10 mg/kg lifibrol (-40%) but not by 5 mg/kg lifibrol (-5%). Triglycerides and free fatty acids were not consistently influenced by lifibrol treatment. In contrast, concentrations of phospholipids in serum, HDL and LDL were lowered by lifibrol treatment. Lifibrol was well tolerated by the pigs and parameters of clinical chemistry, as well as hematological parameters, demonstrated that lifibrol treatment had no side effects on metabolism. This study shows that lifibrol, even in low doses, has a hypocholesterolemic effect and may be useful for therapy of hypercholesterolemia.
