Seed colour in Brassica napus: QTL mapping, candidate genes and associations with quality traits

Abstract
Quantitative trait loci (QTL) contributing to yellow seed colour and content of antinutritive fibre were identified in segregating oilseed rape (Brassica napus) mapping populations ('YE1' and 'YE2') developed from two distinct sources of yellow-seeded oilseed rape crossed with different black-seeded genotypes. Genetic maps were constructed using AFLP and SSR markers, and two corresponding QTL for yellow seed colour were localised in the respective maps, along with a third minor locus in YE2. In both crosses two QTL for seed colour were found to colocalise: The first was a major locus explaining over 60% of the phenotypic variance in seed colour in both cases, while a second, minor locus was also localised at the same map position in the two populatons. In YE2 a further minor gene locus was detected, but not in YE1. QTL and segregation data for seed colour in the two crosses were compared with acid detergent fibre (ADF) content, revealing a causal relationship between the two traits. Potential candidate genes from the flavonoid biosynthesis pathway are now being compared to the QTL positions with a view towards map-based identification of the responsible genes in B. napus.
