Association of Hydrogen Peroxide Accumulation with Expression of PR-1 during Defense of Barley Against the Powdery Mildew Fungus

Abstract

The role of reactive oxygen intermediates (ROI) in the resistance of crop plants against their microbial pathogens is a matter of debate since several years. We here examined tissue-specific accumulation of ROI in different barley lines expressing race-specific resistance gene (Mla12, Mlg) mediated and broad spectrum (mlo5) resistance after inoculation with the barley powdery mildew fungus (Blumeria graminis f.sp. hordei). We found a genotype-specific subcellular accumulation of both superoxide radical anions (O2•¯) and hydrogen peroxide (H2O2). H2O2 accumulation is linked temporally and spatially to papilla formation or the hypersensitive cell death reaction (HR) restricting pathogen growth effectively. Additionally, mRNA accumulation of a pathogenesis-related protein 1 (Hv-PR-1b) and a lipoxygenase 2 (LOX2:Hv:1), which is known to be activated after chemical induction of resistance, were analyzed in near-isogenic barley lines after inoculation with Bgh or after treatment with H2O2, respectively. Expression of these genes is discussed in context of ROI accumulation and induced resistance.
