Abstract:
Preparation technique of BALF might influence differential cell counts. In this study, accelerations of 100, 200, 300, 400, 500 and 600 g and duration of centrifugation (five and ten minutes) were evaluated. Slides were prepared of 15 horses each, either by cytocentrifugation ("glass-cover"-method) or by conventional cell smears. A comparison of cell differentials on cytoslides (100 g for five minutes) with smears was done. The influence of storage at room temperature with or without 40% ethanol and at 4 degreesC with or without 20% phosphate-buffered formalin was tested at 0, 8, 24 and 48 hours. Statistically significant differences between cell differentials were not detected within the slides of either preparation technique. However, significant higher percentages of lymphocytes and lower percentages of foamy alveolar macrophages, neutrophils and mast cells were found on conventional smears in comparison to cytopreparations (100 g and five minutes). Since the clinically especially important neutrophils were detected in highest percentages on cytoslicles which were prepared by spinning the BALF for five minutes at 100 g, these conditions are proposed for standardization of cytologic evaluation of equine BALF.Already after eight hours of storage, absolute and differential cell counts were altered considerably. Storage at 4 degreesC without additive showed the weakest effects.
