Abstract

Thermotolerant and thermophilic actinomycetes were investigated for growth and morphological appearance using a uniform procedure. Therefore reference strains of Actinomadura rubrobrunea, Saccharomonospora viridis, S. caesia, S. glauca, Saccharopolyspora rectivirgula, Streptomyces thermoviolaceus, S. thermovulgaris, Thermoactinomyces thalpophilus, T. sacchari, T. vulgaris, Thermobifida fusca, Thermocrispum municipale, Thermomonospora chromogena and T. curvata were inoculated to six different media (glycerine-arginine-, Difco-actinomycetes-, oats-, R2A-, CASO- and the specifically recommended media from the DSMZ). All plates were incubated at 30, 50 and 55 degrees C for 14 days. Colonies showing the characteristic appearance of actinomycetes were not observed on all media. Often, the colony morphologies were specific for the media used. The colony based identification of the tested strains as an actinomycete was not possible for some strains. It was observed that some of the strains grew into the media but did not produce a colony on the surface. Apparently, none of the tested media was suitable for a general use concerning all thermotolerant and thermophilic actinomycetes.
